Based in Quantico, Virginia, the Behavioral Analysis Unit (B.A.U.) is a subsection of the F.B.I. Called in by local Police departments to assist in solving crimes of a serial and/or extremely violent nature where the perpetrator is unknown (referred to by the Unit as the unknown subject or "unsub" for short), the B.A.U. uses the controversial scientific art of profiling to track and apprehend the unsub. Profiling entails coming up with basic characteristics of the unsub and the victims (referred to as the victimology), using evidence from the case and matching that information to historic precedents and psychological analyses as a means to solve the case. Because of the nature of the work conducted by the B.A.U. - the work being time consuming and psychologically demanding - its members are fiercely loyal to the Unit and to its other members. Also because of the work's overall demanding nature, not many members of the B.A.U. have been able to maintain a happy or stable family life.

Set in modern times, vampires roam the Earth and are humans' mortal enemies. However, a substitute has been found for human blood, a substitute that vampires can consume with the same effect as human blood. This is True Blood. Now humans and vampires can co-exist in harmony, and even form relationships. But there will always be rogue elements on both sides ready to exploit the uneasy truce.

Sydney Bristow (Jennifer Garner) is a young, athletic, college graduate who was recruited her freshman year as a Secret Agent for SD-6, a top-secret branch of the C.I.A. After a few years, after Sydney confides her lifestyle to her boyfriend, the evil head of SD-6, Arvin Sloane (Ron Rifkin), has him killed. Sydney learns that SD-6 is part of a rogue international agency called the Alliance of 12, out to rule the world. She becomes a double agent, working with the real C.I.A. to bring down SD-6 with the assistance of her handler, Michael Vaughn (Michael Vartan), and her estranged father Jack Bristow (Victor Garber), also a double agent. Along the way, Sydney fights various rival agents, rival terrorist groups, and traitors all the while keeping her cloak-and-dagger lifestyle a secret from her friends (season one).

Ethically-challenged attorney Alan Shore, formerly of Young, Frutt & Berluti, settles in at a wealthy and powerful firm focusing on civil cases. With some help from his friend and mentor, veteran attorney Denny Crane, Shore quickly makes his mark winning cases no one would take, often using less than honest methods. In doing so, he develops a rival in his colleague Brad Chase, who has been assigned to the office partly to keep an eye on the increasingly eccentric (and possibly senile) Denny Crane. Though his questionable conduct might make him a few enemies along the way, Alan's not one to be underestimated, nor will he let trivial things like honesty or integrity get in the way of winning a case.

In the Los Angeles office of the Federal Bureau of Investigation, Special Agent Don Eppes and his team investigate critical and baffling crimes with a special edge. That advantage is Don's brother, Charles Eppes, a brilliant universalist mathematician who uses the science of mathematics with its complex equations to ferret out the most tricky criminals. With this team, the forces of evil learn their number is up.

Richard "Rick" Castle is a millionaire playboy who recently killed off his main character when a serial killer starts killing people like he does in his books. He teams up with New York police detective Kate Beckett to solve the case. He finds inspiration in Detective Beckett and starts shadowing her for his next book.

In this modernized version of the Conan Doyle characters, using his detective plots, Sherlock Holmes lives in early 21st century London and acts more cocky towards Scotland Yard's detective inspector Lestrade because he's actually less confident. After the unsolved murder of his wife, Trudy, Adrian Monk develops obsessive-compulsive disorder, which includes his terror of germs and contamination. His condition costs him his job as a prominent homicide detective in the San Francisco Police Department, but he continues to solve crimes with the help of his assistant and his former boss.
