Helen Harris (Kate Hudson) is living the life she's always dreamed of: her career at a top modelling agency is on the rise; she spends her days at fashion shows and her nights at the city's hottest clubs. But her carefree lifestyle comes to a screeching halt when one phone call changes everything. Helen soon finds herself responsible for her sister's children: fifteen-year-old Audrey (Hayden Panettiere), ten-year-old Henry (Spencer Breslin), and five-year-old Sarah (Abigail Breslin). No one doubts that Helen is the coolest aunt in New York City, but what does this glamor girl know about raising kids? The fun begins as Helen goes through the transformation from super-hip to super-mom, but she quickly finds that dancing at 3a.m. doesn't mix with getting kids to school on time, advice that Helen's older sister, Jenny Portman (Joan Cusack), is only too quick to dish out. Along the way, Helen finds support in the most unusual place, with Dan Parker (John Corbett), the handsome young pastor and principal of the kids' new school, and realizes the choice she has to make is between the life she's always loved and the new loves of her life.

Chicago Sun Times copy editor Josie Gellar (25), who was desperate to graduate from perfectionist copy editor to reporter, gets her chance when the goody owner orders the editor to cover the high-school scene by undercover. Josie, who was a frustrated, ridiculed nerd, gets a popular make-over from her drop-out, naturally funny brother Rob Geller. Both siblings find love and joys of youth again. But in Josie's case, it's sensitive bachelor teacher Sam Coulson, who enjoys sophisticated conversation. As the publication deadline approaches, the price of blowing their cover seems ever more daunting, yet inevitable unless she sacrifices her career.

Jake and Brian are friends. They are Jewish and Catholic respectively. They would grow up and become a rabbi and priest. Anna, whom they knew when they were younger, comes back to town a stunning woman. Jake is up to be the head of his synagogue but he is not married which doesn't make his appointment any easier. Jake finds himself attracted to Anna but because she's not Jewish, he can't marry her as it would make his appointment less likely. Brian also finds himself attracted to Anna, but the priesthood doesn't allow that. Their friendship is strained when each learns of the other's feelings for her.

Things get a little awkward when Harry suffers a heart attack at the home of Marin's mother Erica. Left in the care of Erica and his doctor, a love triangle starts to take shape.

Kassie (Jennifer Aniston) and Wally (Jason Bateman) are best friends. Being unlucky in love, Kassie has decided to have a child using artificial insemination. Wally doesn't like this idea, but he isn't capable of admitting to himself, let alone to Kassie, that he's in love with her. At Kassie's artificial insemination party, Wally gets very drunk and spies the sperm donor's sample in the bathroom. Wally was way too drunk to know what he did that night, and Kassie has moved away because she doesn't feel that New York City is a place to raise a child. Now 7 years later, Kassie has moved back with her son Sebastian. While she is looking to get Roland (the sperm donor) more involved in their lives, Wally can't help but notice the many striking similarities that he and Sebastian share.

Sisters Kat and Daisy work along with Jojo at the pizza parlour in Mystic, Connecticut. Kat, shortly off to Yale, finds herself drawn to a local architect she is babysitting for, while her more tearaway sister starts dating a guy from the money side of the tracks. Jojo leaves her man at the altar; she loves him but shies away from commitment. Meanwhile the fame of the pizza continues to spread; it seems to contain something almost ..... mystic.

In Manhattan, the lawyer Liv and the school teacher Emma have been best friends since their childhood. They both are proposed to by their boyfriends on the same day and they plan their wedding parties in Plaza Hotel, using the services of the famous Marion St. Claire. However, due to Marion's secretary's mistake, their weddings are scheduled for the same day. None of them agrees to change the date and they become enemies, trying to sabotage the wedding party of the rival.

In Boulder, Colorado, Grady, who comes from an old money family but who has struck out on his own as a half owner of a fly fishing business, has just unexpectedly passed. Beyond her extreme grief, Gray, his loving fiancée, the two who were to be married in only a couple of days, is placed in a difficult position as they had no assets in their joint names, and without a will she is legally left with nothing of his except the engagement ring, one half of a family heirloom set along with the wedding ring she was about to receive, the engagement ring which his mother, Ellen, would like back. Gray cannot even afford to live in the rental house she and Grady were to live in, and as such, she, at least in the short term, moves into Grady's old room with his two roommates and best friends, Dennis, Grady's quiet, straight-laced business partner, and somewhat clueless Sam, a copywriter of sorts with Celestial Seasonings Tea. The fourth in that buddy friendship is Fritz, a Los Angeles based director of commercials, he who came in for the funeral and who is staying with Dennis, Sam and now Gray for as long as he is in town. Gray has never much liked seemingly self-absorbed Fritz, his current presence which only strengthens that notion as he displays no sense of anything about Grady, let alone grief. In the process of dealing with his estate, Gray discovers some hidden aspects of what was Grady's recent life, both of a financial nature and an associated personal nature. She turns in part to Grady's friends for answers and support, and while Dennis is the one who wishes she turned to in being secretly in love with her, Gray ends up bonding unexpectedly with Fritz as this information answers a lot of questions she had about his and Grady's friendship. Things within this grouping get even more complicated with the arrival into town from Los Angeles of a woman named Maureen and her preschool aged son Mattie, who are somewhat in the center of that hidden issue concerning Grady.

Carmen Lowell is working on the backstage of a play at Yale. When the lead actress, her friend Julia, invites her to Vermont with her to work on a play with a professional cast, she decides to stay with her friends and her pregnant mother. However, she changes her mind after she finds that her friends will all leave for the summer: Lena Kaligaris will travel to a drawing course, Bridget Vreeland, who is in an existential crisis, missing her mother, will travel to an archaeological dig in Turkey, and Tibby Tomko-Rollins is working in a rental and still editing her documentary. Carmen grows a crush on the lead actor Ian, who convinces her to participate in an audition, and she is then invited by the director Bill Kerr to perform the lead female role. Later she finds the truth about the friendship of Julia. The broken-hearted Lena finds that Kostas has just married, and she dates the model of her drawing class. Bridget finds letters addressed to her from her grandmother that her father has hidden, and decides to visit her, discovering information about her mother. Tibby makes love with her boyfriend Brian McBrian and gets scared of a possible pregnancy. In the end, the girlfriends discover that their friendship is real and amounts to more than just a pair of jeans.

They try to forget the whole thing and agree that it means nothing. Ethan, Rachel's close friend, tells her that she'll spend her entire life regretting that if she does nothing.
