On the night of one of their old high school friend's wedding three irresponsible and capricious bridesmaids reunite for one last bachelorette bacchanal in the Big Apple. They unintentionally create a mess of their best friend Becky's wedding dress, before she marries her sweetheart Dale. They attempt to repair the situation by spending the evening before and morning of the wedding desperate to get the dress to Becky on time before the wedding starts, whilst discovering themselves and what they truly want from their lives along the way.

Six New Yorkers juggle love, friendship, and the keenly challenging specter of adulthood. Sam Wexler is a struggling writer who's having a particularly bad day. Violet and her two cohorts attempt to help their "less-fortunate" students at Seven Oaks College - primarily by running a Suicide Prevention Centre and offering their off-beat advice whenever they get a chance. Violet's newest rescue is transfer student, Lily, and Violet wants to teach her how to talk and dress properly, and how to select appropriate men to be interested in. Along their way in helping everybody at the college, the damsels teach the fraternity doofi to hit the books, they get their hearts broken, but then attempt to start an international dance craze.

The film starts with Mindy and Fred ending up in bed together. In the morning Fred is worried that they had unprotected sex during the night and convinces Mindy to get a "morning after pill". However it turns out that it must be followed by a second pill 12 hours later. To make sure that the care free Mindy takes the second pill Fred decides to spend the day with her. It turns out to be an eventful day. Fred learns to know Mindy better while he tries to sort out some other personal challenges.

Dumped by her boyfriend just three weeks before their wedding, Lola enlists her close friends for a series of adventures she hopes will help her come to terms with approaching 30 as a single woman.

Sam has roped his friend Marshall into going on a weekend outing. They are: Hoberg, who is resisting the notion of retirement; Judith Fairfield, Jesse's favorite professor, although for a different reason than his like of Hoberg; Nat, a free spirit who navigates life at the institution on his own terms; undergraduate student Dean, who Jesse sees as a younger more destructive version of himself; and nineteen-year-old undergraduate student Zibby, who is seemingly wise beyond her years and with who Jesse embarks on a relationship despite their sixteen-year-age difference.

Do we ever get what we want? Brian sells mattresses in a warehouse store. His father and older brothers have material success; he wants a child. He's applied to adopt a baby from China. A man who appears homeless seems to be stalking Brian with violent intent. He meets Happy, the daughter of a rich, quirky customer. Adam (Mark Ruffalo) has just reached the 5-year mark in his sex addiction sobriety with help from his sponsor Mike (Tim Robbins). New-comer Neil (Josh Gad) seeks out Adam's help hoping that he'll be his mentor, but Neil doesn't have the same maturity and continues to harass women at work, on the street, and on the subway. Adam has also just met Phoebe (Gwyneth Paltrow), who might be perfect for him, but Adam hasn't been in a relationship since he recognized his addiction to sex, and Phoebe doesn't date addicts. As Adam navigates the romantic relationship waters, Mike struggles connecting to his former drug-addicted son who has just returned home, and Neil develops a relationship with another woman in his sex addicts group, but a platonic friendship might be exactly what he needs.

In San Francisco, after a year's relationship, Tom proposes to Violet; she accepts. She's an experimental psychologist, hoping for a post-doc at Cal. He's a sous chef who runs the kitchen when the chef is away. When Cal falls through and she gets an offer in Ann Arbor, Tom agrees to support the move, turning down a job as chef at a new restaurant. The move requires postponing the wedding. At Michigan, Violet is in her element, but Tom is underemployed and frustrated; he's Stoic for a while, but when two years in Michigan become four, Tom's frustrations boil over, and on the eve of yet another wedding date, they must make a choice. Is there any other alternative?

David sells cars at his father's dealership and, from time to time, visits his grandpa, Joe, at a retirement home. David's a great salesman and he's successful attracting women, one after another. Joe, meanwhile, misses his deceased wife and is done with life. David gives Joe advice on how to get women's attention at the care center, and Joe gives David advice on how to find a lifelong companion. Each dismisses the other's words, then tries them out - David with Julie, a woman he met playing football, and Joe with Edna and with Rose. Failures follow success. Can either figure out how to enjoy life and be themselves?

Iris invites her friend Jack, who's mourning his brother's death, to her father's cottage for some alone time to recuperate, but they discover that Iris' sister Hannah is already there recovering from a recent break-up. One whole bottle of tequila later, Jack and Hannah wake up to find Iris at the door. All three have secrets they're trying to keep and feelings they're trying to sort out.
