Nic and Jules are in a long term, committed, loving but by no means perfect same-sex relationship. Nic, a physician, needs to wield what she believes is control, whereas Jules, under that control, is less self-assured. During their relationship, Jules has floundered in her "nine to five" life, sometimes trying to start a business - always unsuccessfully - or being the stay-at-home mom. She is currently trying to start a landscape design business. They have two teen-aged children, Joni (conceived by Nic) and Laser (by Jules). Although not exact replicas, each offspring does more closely resemble his/her biological mother in temperament. Joni and Laser are also half-siblings, having the same unknown sperm donor father. Shortly after Joni's eighteenth birthday and shortly before she plans to leave the house and head off to college, Laser, only fifteen and underage to do so, pleads with her to try and contact their sperm donor father. Somewhat reluctantly, she does. He is late thirty-something Paul, a co-op farmer and restaurateur. Despite his seemingly successful businesses, Paul has always shirked responsibility, most specifically in his personal life. After Joni and Laser meet with Paul, Nic and Jules learn what their children have done and, although they don't want Paul infiltrating their lives, want to meet him - especially as Joni and Laser seem to want to maintain some sort of relationship with him. As Paul's relationship with the entire family grows (which includes his hiring Jules to design and construct his backyard), they have an effect on what he wants in life. In turn, he affects their family dynamic as well as each person's relationships outside of it.

Six New Yorkers juggle love, friendship, and the keenly challenging specter of adulthood. Sam Wexler is a struggling writer who's having a particularly bad day. When a young boy gets separated from his family on the subway, Sam makes the questionable decision to bring the child back to his apartment and thus begins a rewarding, yet complicated, friendship. Along the way, Helen finds support in the most unusual place, with Dan Parker (John Corbett), the handsome young pastor and principal of the kids' new school, and realizes the choice she has to make is between the life she's always loved and the new loves of her life.

She finds herself pregnant, which throws her plans awry. She bakes phenomenal pies at Joe's diner, listens to old Joe's wisdom, tolerates her sour boss Cal, is friends with Dawn and Becky (her fellow waitresses), and finds a mutual attraction with the new doctor in town. As the pregnancy advances, life with Earl seems less tolerable, a way out less clear, and the affair with the doctor complicated by his marriage. What options does a waitress have?

Jake and Brian are friends. They are Jewish and Catholic respectively. They would grow up and become a rabbi and priest. Anna, whom they knew when they were younger, comes back to town a stunning woman. Jake is up to be the head of his synagogue but he is not married which doesn't make his appointment any easier. Jake finds himself attracted to Anna but because she's not Jewish, he can't marry her as it would make his appointment less likely. Brian also finds himself attracted to Anna, but the priesthood doesn't allow that. Their friendship is strained when each learns of the other's feelings for her.

Kassie (Jennifer Aniston) and Wally (Jason Bateman) are best friends. Being unlucky in love, Kassie has decided to have a child using artificial insemination. Wally doesn't like this idea, but he isn't capable of admitting to himself, let alone to Kassie, that he's in love with her. At Kassie's artificial insemination party, Wally gets very drunk and spies the sperm donor's sample in the bathroom. Wally was way too drunk to know what he did that night, and Kassie has moved away because she doesn't feel that New York City is a place to raise a child. Now 7 years later, Kassie has moved back with her son Sebastian. While she is looking to get Roland (the sperm donor) more involved in their lives, Wally can't help but notice the many striking similarities that he and Sebastian share.

Four women friends: three are wealthy and married plus there's Olivia, a former teacher who's now a maid. The marriages are in various states of health: Franny and Matt are happy and very rich. Christine and David write screenplays together, are remodeling their house, and argue. Jane is angry all the time and Aaron, who's an attentive husband, strikes everyone as gay. Franny sets up Olivia with a friend of hers, Mike, a personal trainer, and Olivia takes him with her to a couple of housecleaning jobs. A benefit dinner for ALS, an awkward guy named Marty whose place Olivia cleans, and a French maid's outfit figure in the story. Is there more to life than its problems?

Becky (Rachel McAdams) is a hard-working morning TV show producer, or at least she was until she got fired. Desperate to get a job, she finally gets an interview with Jerry (Jeff Goldblum) - who is desperate to hire a producer for the struggling show "Daybreak". Becky accepts the job and it proves to be more difficult than even she might be able to handle. She has to fire the sexist co-host, then try to convince egotistical news reporter, Mike Pomeroy (Harrison Ford), to take the job, and then try and get him to actually do the job, properly. And she has to do this while falling for handsome Adam (Patrick Wilson), and trying to save the show from plummeting ratings. Will Becky be able to hold on to her dream job and her sanity?

Although the couple think that this affair will stay in a different state, it brings itself back to their own city and disrupts their personal lives. While the couple still maintain other romances, they cannot help but to continue with their affair.

Everett Stone, who made it on Wall Street, returns home for Christmas with his new girlfriend, Meredith Morton. His ultra-liberal, anti-conventional rustic Connecticut family doesn't exactly warm to the outsider, who despite her best efforts to please the prospective in-laws, looks, sounds, and acts like the conservative bigots they hate, while various Stones have their own problems. Only matriarch Sibyl Stone's unshakable maverick other son Ben gives her a chance, and as Everett won't actively turn against either, she feels more supported by him and a weird romance blossoms. Feeling beleaguered, Meredith calls in help from her easygoing sister Julie, who proves no help to her but soon develops a chemistry with Everett, so everything may now shift if they dare follow their hearts.

After graduating from Montclair State, New Jersey Girl Annie can't make up her mind about what to do with her life. After saving a little boy from being run over in the park, she is quickly employed as a nanny for a rich Upper East Side couple. Mr X is occupied with his business, Mrs X loves shopping, and neither really likes to spend time with their little boy Grayer. Annie quickly learns that she has more than her hands full taking care of him. Her busy schedule doesn't give her much spare time. Mrs X fired her last nanny because she was dating and that gives Annie problems when Harvard Hottie who lives in the same building asks her out on a date.

Then she meets Adam (Paul Rudd) , an attractive, spontaneously funny, single, 29-year-old actor at an audition. She successfully casts him, which revives the show's ratings. She also dates him, but her pathological insecurity, focused on their age difference, compromises the relationship. That culminates when she suspects him of infidelity with the show's star, and the studio gives those two their own sitcom.

Iris invites her friend Jack, who's mourning his brother's death, to her father's cottage for some alone time to recuperate, but they discover that Iris' sister Hannah is already there recovering from a recent break-up. One whole bottle of tequila later, Jack and Hannah wake up to find Iris at the door. All three have secrets they're trying to keep and feelings they're trying to sort out.

Peter is a composer and a likable sad sack who's devastated when his girlfriend of five years, Sarah Marshall, the star of a cheesy CSI-style crime show, dumps him. He weeps, he rails, he mopes. Finally, his step-brother Brian suggests a vacation, so Peter heads for a resort on Oahu where, as he's checking in, he sees Sarah and her new beau, Aldous, a perverse English rocker. The weeping and moping start again, until Peter is rescued by Rachel, a thoughtful hotel clerk who invites him to a luau and to hang out. Although he constantly runs into Sarah and Aldous, Peter starts to come alive again. Will Sarah realize what she's lost, and what about Rachel?

Henry Roth: obsessive-compulsive, somewhat misanthropic, a writer of children's books. His illustrator and only friend, Rudy, dies after a fabulously successful collaboration on "Marty, the Beaver." Henry is under contract to produce another Marty book for Christmas sales. His publisher, Arthur Planck, assigns penniless, lovelorn illustrator Lucy Reilly to work with Henry. She's sought by her ex-boyfriend Jeremy, who dumped her two years ago but shows up apologetic, having dedicated his new book to her. She and Henry go to a house on the shore to work. Will love bloom amid the rocks, or is Henry a bump on Lucy's road to Jeremy? Rudy's voice, from the grave, gives Henry counsel.
